Research
- Palate bone formation and the role of the hypoxia pathway

- The central hypothesis is that the Hif1α/VEGFa response to hypoxia during palate bone development is a conserved pathway and mediators of this pathway can be identified as potential therapeutic targets for maxillary hypoplasia. This pathway has been interrogated in orthopedic literature involving long bones but has not been investigated in the craniofacial skeleton. Current treatment for maxillary hypoplasia requires surgical correction, and often autogenous bone grafting (ABG). ABG is limited in tissue supply and associated with significant cost and morbidity. The alternative to ABG, bone morphogenetic protein 2 (BMP2), is FDA approved in adults, but is not available to children due to concerns for life-threatening inflammation and oncogenic potential. Regenerative approaches to treat bone defects in children avoiding ABG are critically needed. We are investigating the role of the Hif1α/VEGFa pathway in bone formation using cell cultures to identify downstream mediators that can be delivered in a hydrogel to induce osteoblast commitment and bone formation in vitro and in vivo. This research is a critical step in the development of a novel therapeutic option for children with maxillary hypoplasia.
